Under supervision and direction in a line to the Clinical Lab Supervisor and the Technical Director of the Immunopathology, incumbent is responsible for performing all assays for autoantibody detection in patients with suspected or actual Wegener's granulomatosis or other related vasculities. The diagnostic information is used by clinicians for treatment and patient care management.

Assists in the day-to-day operations and overseeing of the ANCA laboratory of the Immunopathology Unit. Trains new technologists on proper procedures and techniques, evaluates new technology for clinical application and assists in updating procedure manuals for laboratory inspections and accreditation such JCAHO.

Functions as principal technical specialist for the ANCA laboratory, provides instruction in special techniques, methodology and the reading and interpreting of test results to students, residents; provides all of the above except the reading and interpreting of test results to the pathologists.

Responsible for performing indirect immunofluorescence and ELISA (immunoassay) for detection of circulating autoantibodies such as ANCA (anti-neutrophil cytoplasmic antibodies) which are associated with life threatening diseases and syndromes that involve the lung and kidney. Responsible for quality control of reagents including antibodies.

Utilizes phlebotomy techniques for obtaining normal blood donors for the isolation of neutrophils. Prepares cytosmears of the isolated neutrophils to be used for indirect immunofluorescence.

Isolates proteinase-3 (p-29), one of the antigens in the immunoassay (ELISA), by using affinity column chromatography techniques.

Assessions samples, logs data, files reports and orders supplies.

Coats microtiter plates with antigens, blocks plates and performs the indirect or direct ELISA assay. Washes plates, develops in enzyme substrate and reads optical densities using ELISA plate reader. Aliquots sera and freezes them after assay is performed.

Additional duties include operation and maintenance of all laboratory equipment as well as development and implementation of new and improved techniques and protocols to ensure high quality results. Additionally, the technologist actively participates in teaching and training of pathology residents and fellows. Instructs pathology residents in the reading and interpretation of test results.

The technologist also maintains and updates the laboratory manual in accordance with the JCAHO regulations and implements hospital safety standards with the help of the Technical Director.

Workflow and workload of diagnostic specimens may require working Friday evenings

Maintains quality control logs on a variety of equipment.

BS or BA. in Medical Technology, Clinical Laboratory Science or Chemical, Physical, or Biological Science and with MT ASCP Certification and 3-5 years experience .

Familiarity with immunology, computers, and the metric system. Knowledge of chemistry; familiarity with the concepts of molarity, normality, molecular weight, formula weight of chemicals. Familiarity with how to make dilutions from stock solutions. Knowledge of good laboratory technique. Must be able to do mathematical calculations, and read/write English. Must know proper handling procedure of chemicals.

Effective interpersonal and communication skills, attention to detail. Must be organized and capable of following protocols and have ability to devise or adapt techniques if needed to achieve desired goals. Supervisory skills are needed to instruct technologist or residents. Ability to recognize novel or emergency situations and respond appropriately. Good hand and eye coordination and manual dexterity. Ability to work well with others.
